We currently have two tandem mass spectrometers operated in conjunction with our nano UPLC systems and autosamplers which are dedicated to protein identification, quantitation and PTM identification. The Orbitrap Elite is dedicated to proteomics analysis and serves as our workhorse instrument while the older LTQ is used primarily for method development and non-standard analyses.

  • Thermo Fisher Orbitrap Elite with Waters NanoAcuity UPLC
    The Orbitrap Elite is a hybrid instrument combining a linear ion trap quadrupole mass filter with a high resolution orbitrap mass analyzer which can be operated at resolutions up to 240,000. The instrument uses a nanoscale ionization source interfaced with the NanoAcquity which is operated at a nominal flow rate of 250 nanoliters per minute. This configuration offers rapid, sensitive, highly accurate and robust technology for the identification of proteins from proteolytic digests of gel-based or solution based samples.

    These instruments are used for quantitation, PTM identification protein identification. The high resolution and mass accuracy make this an ideal instrument for quantitation from isotopicaly and isobaricaly labeled proteins. We generally inject 10 ul of 20 ul final sample volume using the NanoAcquity Sample Manager although we can inject as much as 50 ul.

  • Shimadzu HPLC and Fraction Collector
    This HPLC is used primarily for reversed phase chromatographic separation and fraction collection of proteomics samples destined for Orbitrap analysis. The HPLC can accommodate different buffers for the identification of small molecules upon request. (Our capacity for this is limited by the age of this instrumentation.)

  • Thermo Fisher LTQ with Dionex Ultimate 3000 Rapid Separation LC (RSLCnano)
    The LTQ is a linear ion trap quadrupole mass spectrometer purchased in 2005. We use this instrument for chromatographic method development as well as non-standard analysis of samples not well suited for the Orbitrap. The instrument can be interfaced to a nanoscale ionization source for use with the RSLCnano as well as being interfaced with the Shimadzu HPLC (manual injections) operated at micro and analytical flow rates.
